Application
The enzyme has been used along with amyloglucosidase for the in vitro digestion of starch in food samples.[1] Pancreatin from Sigma has been used for in vitro pepsin-pancreatin digestion assays while estimating of lower gut digestibility using rumen feed residues. It has also been used along with pepsin to simulate in vitro gastric and ileal digestion of raw materials in pigs.
Pancreatin is a mixture of several digestive enzymes produced by the exocrine cells of the porcine pancreas. It is a broad-spectrum protease composed of amylase, trypsin, lipase, ribonuclease and protease. It is used for in vitro digestibility analysis[2]. Product P7545 has been used to test the sensitivities of cellulolytic bacteria inhibitors[3].
Biochem/physiol Actions
Due to the enzymatic components such as trypsin, amylase and lipase, pancreatin hydrolyzes proteins, starch and fats. Pancreatin will convert not less than 25 times its weight of potato starch into soluble carbohydrates in 5 minutes in water at 40 °C, will digest not less than 25 times its weight of casein in 60 minutes at pH7.5 at 40 °C and will release not less than 2 microequivalents of acid per min per mg pancreatin from olive oil at pH9.0 at 37 °C.

This product has not been tested in-house for the application of assessing the effect of enzymes in pancreatin on fungus and fungal cell walls, specifically with lipase. A recommended concentration has not been established. Conducting a literature search may provide guidance on this matter.

This product meets 8X the minimum USP specifications. According to the USP monograph for Pancreatin, the 1X minimum specification is 25 USP units amylase, 2 USP units lipase, and 25 USP units protease per milligram. The calculated protease activity for the 8X preparation then, would be at least 200 units per milligram. This is a calculated value based on the USP monograph and are not reported separately on the Certificate of Analysis.

Hi, what is the dissovled soultion? It can't dissolved in water or 1 mM HCl

This product is not tested for solubility. For assay purposes. it is dissolved at 40 mg/3mL PBS, pH 6.8. Trituration is required for 5 - 10 minutes. Historically, this product has also been dissolved in water at 20 mg/ml. with heating at 37 °C. This method yields a hazy solution.

The specific activity of this pancreatin is specified to be at least 8 x the minimum USP standard.
The USP defines a minimum specification of a 1 X Pancreatin as follows:
- 25 USP protease units per mg.
- 25 USP amylase units per mg.
- 2 USP lipase units per mg.Therefore, activity will be 8 times that.
