Application

Pancreatin from porcine pancreas has been used to assess the treatment of steatorrhea by lipase supplementation therapy, to investigate treatment options for pancreatic diabetes in patients experiencing the decompensated stage of chronic pancreatitis, and to safely and effectively remove formalin-fixed tissues from arterial grafts without leading to structural damage or loss in fiber integrity.

Biochem/physiol Actions

Pancreatin contains enzymatic components including trypsin, amylase and lipase, ribonuclease, and protease, produced by the exocrine cells of the porcine pancreas. This combination of enzymes allows it to hydrolyze proteins, starch and fats. Pancreatin will convert not less than 25 times its weight of potato starch into soluble carbohydrates in 5 minutes in water at 40°C, will digest not less than 25 times its weight of casein in 60 minutes at pH 7.5 at 40°C and will release not less than microequivalents of acid per min per mg pancreatin from olive oil at pH 9.0 at 37°C.

    1. This product is tested according to the USP monograph which determines amylase, lipase, and protease activity only. The total protein content and trypsin mass fraction are not determined for this material. Each lot is released as meeting USP specifications. Individual enzyme activities are not reported.

    1. The specific activity of the pancreatin is specified in multiples of the minimum USP standard (USP 24, p. 1254). The USP defines a minimum specification of 1X Pancreatin as follows:
      25 USP protease units per mg.
      25 USP amylase units per mg.
      2 USP lipase units per mg.

      For the 4 × USP the above values are multiplied by four. The product Certificate of Analysis confirms that this specification has been met, but no activity values are reported. The USP defines units as:
      One USP Unit of amylase activity is contained in the amount of pancreatin that decomposes starch at an initial rate such that 0.16 µ Eq of glycosidic linkage is hydrolyzed per minute under the conditions of the Assay for amylase activity.

      One USP Unit of lipase activity is contained in the amount of pancreatin that liberates 1.0 µ Eq of acid per minute at a pH of 9.0 and 37° under the conditions of the Assay for lipase activity.

      One USP Unit of protease activity is contained in the amount of pancreatin that under the conditions of the Assay for protease activity hydrolyzes casein at an initial rate such that there is liberated per minute an amount of peptides not precipitated by trichloroacetic acid that gives the same absorbance at 280 nm as 15 nmol of tyrosine.

  3. What are the USP/mg for pancreatin (P3292)?

    1. This product meets 4X the minimum USP specifications. According to the USP monograph for Pancreatin, the 1X minimum specification is 25 USP units amylase, 2 USP units lipase, and 25 USP units protease per milligram. The calculated protease activity for the 4X preparation then, would be at least 100 units per milligram. This is a calculated value based on the USP monograph and are not reported separately on the Certificate of Analysis.
