一般描述

透明质酸酶广泛存在于自然界。在人体中,它存在于不少器官和体液中。透明质酸酶见于病毒、噬菌体、真菌和细菌的分泌物。它还存在于线虫和蚂蟥的分泌物中。蛇和蝎子的毒液中会产生透明质酸酶。蜜蜂、蜘蛛、黄蜂、大黄蜂、毛虫、鱼类和蜥蜴也会分泌透明质酸酶。

应用

透明质酸酶(来自Streptomyces hyalurolyticus)已用于:
  • 制备DMEM/F-12(杜氏改良Eagle培养基/营养混合物F-12)培养基,用于从分离肿瘤组织中分离和纯化单细胞
  • 作为消化液组分,用于从睾丸组织获得生殖干细胞

生化/生理作用

透明质酸裂解酶在β-D-GlcNAc-(1→4)-β-D-GlcA键上切割透明质酸,得到3-(4-脱氧β-D-葡萄糖-4-烯基糖基)-N-乙酰基-D-葡糖胺四糖和六糖。与其他透明质酸酶不同,该酶对透明质酸具有特异性,并且对软骨素和硫酸软骨素无活性。
透明质酸酶属于内切糖苷酶,通过配合β-葡糖醛酸酶和β-N-乙酰氨基葡萄糖苷酶等外切糖苷酶完成其内切功能。透明质酸酶调节透明质酸的周转代谢。

单位定义

在 37 ºC、pH 5.35 条件下,一单位每分钟可降解 0.75 μg 多糖透明质酸(通过比浊吸光度测量(λ: 600nm),与 BSA 复合 45 分钟后测量)。

To measure hyaluronidase activity, a turbidimetric determination assay is used at 600 nm. One unit of hyaluronidase activity will cause a change in absorbance of 0.330 per minute at pH 5.35 at 37 °C.
