一般描述

透明质酸酶是一种透明质酸代谢酶。[1]它发现于蛇、蜜蜂和膜翅目的毒液中。透明质酸酶可降解透明质酸(HA),从而增强膜的通透性、降低粘度并增加组织的通透性。它可增强局麻药的扩散,减小心肌梗塞区域的尺寸,提高抗癌药的效果,并防止了自发分娩时的会阴部创伤。[2]

应用

来自绵羊睾丸的透明质酸酶已被用于:
  • 流式细胞术[3]
  • 分离假定的精原干细胞(SSC)[4]
  • 研究磁刺激对磁性水凝胶性质的影响[5]
  • 在免疫组化中观察II型胶原的分布[6][7]

生化/生理作用

这些酶可随机裂解透明质酸、软骨素和硫酸软骨素中的 β-N-乙酰己糖胺-(1→4) 糖苷键。

To measure hyaluronidase activity, a turbidimetric determination assay is used at 600 nm. One unit of hyaluronidase activity will cause a change in absorbance of 0.330 per minute at pH 5.35 at 37 °C.

    1. This enzyme is soluble at 1 mg/mL in 20 mM sodium phosphate buffer (pH 7.0), with 77 mM sodium chloride and 0.1 mg/mL BSA at 1 mg/mL. For assay purposes, this product is prepared at 0.3 mg/mL solution in Phosphate Buffer. Heating may be required, Please see the link below to review the full assay protocol:
