生化/生理作用

GRP78 / BiP水平在阿尔茨海默′病患者大脑中升高,研究发现GRP78 / BiP表达的降低与早老素1(PS-1)的错义突变(missense mutation)有关。
GRP78(78 kDa 葡萄糖调节蛋白)是维持细胞活力所需的热休克蛋白 Hsp70 成员。它是一种位于内质网 (ER) 的Ca2+-结合分子伴侣。在新合的成膜结合或分泌蛋白的正确糖基化、折叠和组装中,它具有关键作用。该蛋白还有助于保留不正确折叠的突变或存在缺陷的蛋白质,并防止此类蛋白质从细胞质易位至内质网室。该蛋白质可被氧化应激、化学毒性、糖基化和缺氧等条件诱导。它在维持细胞稳态和预防细胞凋亡中具有至关重要的作用,并可作为低血糖的生物标志物。对于暴露于谷氨酸外毒性和氧化应激的神经元,它可提供神经保护功能。
