Serine/threonine-protein kinase RIO2 (RIOK2) is a nonribosomal factor required for normal ribosomal RNA biogenesis and cell cycle progression. It is necessary for processing of 20S pre-rRNA to the 18S rRNA in 40S ribosomal subunit synthesis. RIOK2 is also a ribosomal assembly factor that prevents premature translation initiation on the small (40S) subunit. It is needed for proper mitotic progression, regulates metaphase-anaphase transition and is a substrate of polo-like kinase 1 (Plk1) which is phosphorylated by Plk1 at the Ser-335, Ser-380 and Ser-548 residues.
