Biochem/physiol Actions
DMOGは細胞浸透性プロリル-4-ヒドロキシラーゼインヒビターで、HIF(低酸素誘導因子)をアップレギュレートします。
DMOGは細胞透過性のプロリル-4-ヒドロキシラーゼ阻害剤であり、HIF(低酸素誘導因子)をアップレギュレーションします。HIF-1αサブユニットのタンパク質レベルはプロリルアスパラギニルヒドロキシラーゼ(PAH)により転写後調節されています。PAH活性が抑制されると、内在性HIF-1αの濃度が増加します。DMOGはプロリルヒドロキシラーゼドメイン含有タンパク質(PHDおよびHIF-PH)の細胞透過性競合阻害剤です。NGFを欠乏させた培養細胞では、糖代謝の維持を通じた神経保護作用を有することが見いだされています。また、DMOGはウサギ虚血再灌流モデルで心筋障害を軽減しています。DMOGは従来の阻害剤である4-フェニルピリジン-2,5-ジカルボン酸(製品番号R395889:シグマ アルドリッチレアケミカルライブラリー)よりも強力です。IC50 は5.18 μMです。
