Biochem/physiol Actions

BRAFは、成長因子の刺激でRasファミリー分子により活性化されるRAFファミリーの一員です。細胞において観察される成長因子シグナルに応答したcRafとBRAFの協同作用は、活性型Rasが引き起こすcRafとBRAFのヘテロ二量体化によって説明することができます。BRAF遺伝子の活性化変異はヒト悪性黒色腫でかなりの割合を占めており、一部の大腸癌でも認められます。これらの圧倒的多数において、BRAFの活性化領域にある599番目の残基がバリンからグルタミン酸に変異しています。
