SYBR® Green Iは、核酸の検出に使用される、独自の非対称シアニン染料です。N-アルキル化ベンゾチアゾリウムまたはベンゾオキサゾリウム環系からなり、モノメチン橋によってピリジニウムまたはキノリニウム環系に結合されます。[1]SYBR Green IはdsDNAの副溝と結合し、480 nmの波長で励起されます。ピーク蛍光発光は520 nmです。[2]

アプリケーション

SYBR® Green I 核酸ゲル染色は、次の用途で使用されています：
  • dsDNAの定量[3]
  • ポリメラーゼ連鎖反応（PCR）におけるDNAの染色[4]
  • コメットアッセイ法[5]
  • 精子膜完全性の評価[6]
  • ループ介在等温増幅（LAMP）により増幅したDNAの目視検査[7]
  • フローサイトメトリーにおける蛍光色素として[8]
  • 逆転写されたcDNAの染色のためのリアルタイム定量的逆転写ポリメラーゼ連鎖反応（qRT-PCR）[8]

特徴および利点

  • アガロースゲルまたはポリアクリルアミドゲル中で電気泳動後のdsDNAの染色に用いる、高感度の色素
  • プレ洗浄なしで、アガロース/ホルムアルデヒドおよびポリアクリルアミド/ウレア変性ゲル中の ssDNA および RNA の検出にも使用できます
  • エイムス試験において、臭化エチジウムよりも変異原性が低くなっています
  • オリゴヌクレオチドに対し、臭化エチジウムよりも 50 ～ 100 倍高感度です
  • 少量のDNAにより多くのアプリケーションに有用です
  • SYBR®Green IがDNAに結合しても、Hind IIIおよびEcoR Iなどの多くの一般的な制限エンドヌクレアーゼの活性を阻害しません
  • ゲル内消化およびライゲーション技術によるこの染色液の除去は不要です
  • SYBR Green Iは、より環境に優しい、染色用臭化エチジウムの代替品です

保管および安定性

本製品は -20°Cで保管してください。希釈した染色液は、遮光して、2 ～ 8°Cで数週間、または、室温で 3 ～ 4 日間保管できます。

    1. This product is a 10,000X concentrated solution suitable to prepare 10L of staining solution for in-gel staining of DNA and RNA. The specific concentration is considered proprietary. Published working concentrations for solutions of Sybr Green I range from 0.5 - 1.0 ug/ml. The molecular weight as published by Chebi, is 509.74 g/mol.     1. Product S9430, SYBR® Green I nucleic acid gel stain, will poorly stain DNA containing deaza-G modified nucleotides in place of guanisine.  A way around this issue would be to use a mixture of deaza-G modified nucleotides with guanisine.  This will allow for better staining.
