Descrizione generale

Monocyte chemoattractant protein-1/monocyte chemotactic and activating factor (MCP-1/MCAF), also called the C-C motif chemokine ligand 2 (CCL2),[1] is an inflammatory cytokine.[2] The CCL2 gene is mapped to human chromosome 17q12.[1] CCL2 is present in epithelial, endothelial, mesangial and microglial cells.[3] Structurally, it dimerizes and has four regions of the ß-sheet.[3] Recombinant human MCP-1 is a 8.6 kDa protein containing 76 amino acid residues including the four highly conserved cysteine residues.[3] The MCP family cross-reacts across species.

Applicazioni

Monocyte chemoattractant protein-1/monocyte chemotactic and activating factor (MCP-1/MCAF) has been used:
  • as a chemokine supplement in the alpha minimal essential medium for culturing neurons to test its effect on JmjC histone demethylase (Jmjd2a) mRNA[2]
  • as a component of Dulbecco′s Modified Eagle (DMEM) medium, as a positive chemoattractant control in HeLa cells for migration assay[4]
  • as a chemoattractant to induced macrophage chemotaxis[5]

Azioni biochim/fisiol

Monocyte chemoattractant protein-1/monocyte chemotactic and activating factor (MCP-1/MCAF) acts as a human immunodeficiency virus 1 (HIV-1) suppressor.[3] The MCP-1 acts as a chemoattractant for T cells and natural killer (NK) cells.[1] It also plays a key role in the migration and infiltration of macrophages.[3] CCL2 shows under expression in high-grade ovarian serous carcinomas (HGOSCs) and is expressed less in epithelial ovarian cancer (EOC).[1]
Monocyte chemoattractant protein-1/monocyte chemotactic and activating factor (MCP-1/MCAF) is a member of CC chemokine family. It triggers the activation of monocytes by triggering degranulation as well as respiratory burst activity in monocytes. It facilitates monocytes in inhibiting or enhancing the growth of specific brain tumor cell lines. It is found to be overexpressed in cases of atherosclerosis, idiopathic pulmonary fibrosis, rheumatoid arthritis, and delayed hypersensitivity reactions. It is also implicated in Kawasaki disease.[6]

Ricostituzione

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a stabilizer (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
