凝血酶(活化因子IIa)是一种重要的凝血启动子,可控制可溶性纤维蛋白原向不溶性活性纤维蛋白链的转化。凝血酶是一种凝血蛋白和一种丝氨酸蛋白酶(EC 3.4.21.5),可催化与凝血相关的多种反应。它主要与凝血有关。
生化/生理作用
凝血酶可触发XI因子、V因子、XIII因子和VIII因子。凝血酶通过活化血小板上的蛋白酶活化受体来促进血小板的活化。它还会诱发炎症。由于其高蛋白水解特异性,凝血酶已成为一种重要的生化蛋白。凝血酶切割位点(Leu-Val-Pro-Arg-Gly-Ser)被广泛用于重组融合蛋白构建体的接头区域。在纯化融合蛋白后,凝血酶可被用于在切割位点的精氨酸和甘氨酸残基之间进行切割,从而高特异性的从目的蛋白中高效去除纯化标签。

从由 20 mM Bis-Tris、150 mM NaCl 和 0.1% PEG 8000、pH 6.5 组成的缓冲液冻干。
重構
可以通过添加 1 mL 缓冲盐水,pH 7.4(例如 25 mM HEPES,150 mM NaCl,0.1% PEG 8000,pH 7.4)来制备工作储备溶液。应在含有合适稳定剂(例如 0.1%-1% Prionex、BSA 或 PEG)的缓冲液中进一步稀释。
