Hämoglobin (Hb) ist ein eisenhaltiges Metalloprotein, das als wichtigste Methode zum Sauerstofftransport in Wirbeltieren dient. Hämoglobin kommt hauptsächlich in Erythrozyten[1] vor, wo es 97 % des Trockengehalts der Zelle ausmacht. Hämoglobin ist auch in anderen Geweben anzutreffen, wo es als Antioxidans dient. In vielen Krankheiten wie Anämie und Polyzythämie kommt es zu Änderungen der Hämoglobinwerte im Blut.[2][3]

Eignung
Eignet sich zum Nachweis von Hämoglobin in einer Vielzahl von Proben wie Blut, Serum, Plasma und Urin. Dieses Kit kann zur Untersuchung der Auswirkungen verschiedener Verbindungen auf den Stoffwechsel und die Konzentration von Hämoglobin eingesetzt werden.
Prinzip
Dieser Assay basiert auf der optimierten Triton™[NP1]/NaOH-Methode, bei der Hämoglobin zu einem kolorimetrischen Produkt umgewandelt und bei 400 nm gemessen wird. Dieses Assay hat bei Verwendung einer 96-Well-Mikrotiterplatte einen linearen Detektionsbereich zwischen 0,9 und 200 mg/dL.

Is this a hemolysis kit? Certain capmounds are used to detect compatibility with human blood; is this used to detect the amount of free hemoglobin? Can this be used to find whether our compound has cytotoxicity towards RBC?

The assay kit can be used to detect free Hemoglobin. It is not suitable for use in the detection of substances that are cytotoxic to red blood cells.

How can I determine the shelf life / expiration / retest date of this product?

If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

How is shipping temperature determined? And how is it related to the product storage temperature?

Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. Is there stability information available for the Reagent (MAK115A) and Calibrator (MAK115B) included in the hemoglobin assay kit, especially after they are opened?

The recommended storage for both components is at 2-8 degrees Celsius when not in use. Cycling the components between refrigerated and room temperature may decrease shelf life, but there is no predicted rate of degradation. For prolonged shelf life with frequent use, aliquoting the reagents into smaller tubes for individual use is advised.
