Anwendung
Carbonanhydrase von bovinen Erythrozyten (BCA) wird in Studien eingesetzt, die die Auswirkungen des Entfernens des enzymgebundenen Metalions Zn2+ auf das Aggregationsverhalten des Enzyms untersuchen. Das Entfernen des Metalions durch einen Chelatbildner wie beispielsweise EDTA verbessert die Neigung des Enzyms, den geschmolzenen, kugelförmigen Zustand einzunehmen. In diesem Zustand bindet sich das Enzym an das Chaperon-ähnliche α-Crystallin und verhindert die Aggregation.[1] Das von Sigma vertriebene Enzym wurde zum Einsatz mit elektrochemischen Messwertaufnehmern immobilisiert, um ein analytisches Instrument zur Messung von gelöstem CO2 zu entwickeln.[2] Es wird außerdem für die thermodynamische Analyse von Konformationsänderungen in BCA eingesetzt.[3]
Biochem./physiol. Wirkung
Carboanhydrase ist ein Zink-Metalloenzym mit einem Molekulargewicht von ca. 30.000 Da. Das Enzym katalysiert die Hydration von Kohlendioxid zu Carbonsäure. Es ist an wichtigen physiologischen und pathologischen Prozessen beteiligt, z. B. pH- und CO2-Homöostase, Transport von Bicarbonat und CO2, biosynthetischen Reaktionen, Knochenresorption, Kalzifikation und Tumorigenität. Dieses Enzym ist daher ein wichtiges Ziel für Hemmstoffe und hat klinische Anwendungen für verschiedene Pathologien wie Glaukom, Epilepsie und Parkinson-Krankheit.[4]

Eine Wilbur-Anderson (W-A-)Einheit bewirkt eine Senkung des pH-Werts von 0,02 M Trizma-Puffer von 8,3 auf 6,3 pro Minute bei 0 °C. (Eine W-A-Einheit entspricht im Wesentlichen einer Roughton-Booth-Einheit.)
