Derived from the peripheral blood of a 1 year old male with acute monocytic leukaemia. THP-1 cells have Fc and C3b receptors and lack surface and cytoplasmic immunoglobulins. These cells also stain positive for α-napthhyl butyrate esterase, produce lysozymes and are phagocytic (both latex beads and sensitised erythrocytes). THP-1 cells can also restore the response of purified T lymphocytes to Concanavlin A, show increased CO2 production on phagocytosis and can be differentiated into macrophage-like cells using for example DMSO.

THP 1 cell line human has been used to study:
  • bactericidal activity of moxifloxacin against Staphylococcus aureus[1]
  • the effect of lipopolysaccharide-mediated inflammatory response in murine versus human cell lines[2]
  • the effects of purine nucleosides on the in vitro growth of Cryptosporidium parvum[3]
  • the change in gene expression after infection with Mycobacterium tuberculosis[4]
  • the induction of cytokines in the cell line in presence of antigens obtained from Clostridium difficile[5]

Rutina de subcultivo

If starting from a frozen ampoule the cryoprotectant should be removed. Add thawed cells to a conical based centrifuge tube, e.g. 15ml tube, slowly add 4 ml of culture medium to the tube. Take a sample of the cell suspension, e.g. 100μl, to count cells. Centrifuge the cell suspension at low speed i.e. 100 - 150 x g for a maximum of 5 minutes. Remove medium and resuspend the cell pellet at a density of 3 - 5 x 100,000 cells/ml in fresh medium containing 20% serum. Incubate flask at 37°C; 5 - 7% CO2. Check daily. Keep flask in a vertical position until the cells reach the exponential phase of growth. This can take up to 7 days. Once the culture is established the serum concentration can be reduced to 10%. To keep the cells in exponential growth, maintain cultures between 3-8x100,000 cells/ml. Requires 5% DMSO and 95% fetal bovine serum (FBS) as cryoprotectant. Growing orders are recommended due to difficulties that can be experienced during the initial start-up of this cell line. Replacements will be charged at full cost where claims cannot be substantiated.

  1. Is it possible to provide the differentiation protocol of your THP1 cells into macrophage ?

  2. the word DNA , in the cataloge number (88081201-DNA-5UG) is a bit confusing. can you confirm that it is the frozen cell line but not the frozen DNA extracted from this cell line. Thank you.

    1. For the ECACC cell line products there are three possible product options. SKU designations of -1VL are the frozen cell line stocks. SKU designations of -DNA or -RNA are products extracted from the representative cell line followed by the quantity of each package - in this case, 5ug.

  3. Good Afternoon, What is meant by this cell line being supplied as a 'growing culture' as opposed to a frozen ampule? And what are the culturing protocols after they are received?

    1. Because this is a difficult cell line to resuscitate from frozen, if the customer is located in the European Union, ECACC recommends sending these cells live in culture rather than frozen. You can request it to be sent frozen and likely it must be sent frozen if it is being shipped outside of the European Union.

  4. How do I handle Cell Line frozen cells upon arrival?

    1. Upon receipt, frozen ampules should be transferred directly to vapor phase liquid nitrogen without delay (-135°C) or liquid phase liquid nitrogen if vapor phase is not available. DO NOT use a -80°C freezer as an alternative; this will result in loss of viability.

  6. What medium should I use when I thaw the Cell Line cells?

    1. The medium for each cell line is listed on the product page. It is specific to each cell line. If not visible on the Sigma-Aldrich website product page, the HPA website also contains the same information (www.hpacultures.org.uk). The product number for the cell line is the same on either site.

  7. How many Cell Line cells are in the vial?

    1. Each vial of cells contains 2-3 x 106 cells in 1 ml of freezing media. This is in a 1.8 mL ampule.  For exceptionally large cells, counts may decrease. Suspension cells, generally smaller cells, may contain as many as 4-5 x 106 cells/vial to assure optimal viability upon thaw.

  10. Why is Vapor Phase Liquid Nitrogen preferred for storage of Cell Line cells?

    1. If ampules are immersed into liquid phase of liquid nitrogen, it increases the risk of the liquid seeping into the vial. This could lead to problems of cross-contamination and increased risk of the ampule exploding when thawed.
