Clone 1J17 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Bromodomain-containing protein 4 (Brd4). it targets an epitope within 24 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the C-terminal region of human Bromodomain-containing protein 4 (Brd4).

Anwendung

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Brd4 in Jurkat cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Brd4 in HeLa cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound Brd4 with a KD of 2.1 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Brd4 in HeLa cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Brd4 in human small intestine tissue sections.

Zielbeschreibung

Bromodomain-containing protein 4 (UniProt: O60885; also known as Protein HUNK1, BRD4) is encoded by the BRD4 (also known as HUNK1) gene (Gene ID: 23476) in human. Brd4 is a ubiquitously expressed chromatin reader protein that recognizes and binds acetylated histones and plays a key role in transmission of epigenetic memory across cell divisions and transcription regulation. It remains associated with acetylated chromatin throughout the entire cell cycle and provides epigenetic memory for postmitotic G1 gene transcription by preserving acetylated chromatin status and maintaining high-order chromatin structure. Brd4 contains two bromo domains (aa 75-147 and 368-440) that mediate specific binding to acetylated histones via Asn 140 and Asn 143, respectively. The first bromo domain is shown to have high affinity for acetylated histone H4 tail, whereas the second bromo domain recognizes multiply acetylated marks in histone H3. It also contains one NET domain (aa 600-682) that mediates interaction with a number of chromatin proteins involved in transcription regulation. Brd4 is released from chromatin upon deacetylation of histones that can be triggered by different signals such as activation of the JNK pathway or nocodazole treatment. During interphase, it is reported to play a key role in regulating the transcription of signal-inducible genes by associating with the P-TEFb complex and recruiting it to promoters. It is also reported to recognize and bind acetylated RELA, leading to further recruitment of the P-TEFb complex and subsequent activation of NF-kB. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physikalische Form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Rekonstituierung

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Lagerung und Haltbarkeit

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
