Anwendung

Lektin aus Phaseolus vulgaris (Gartenbohnen) hat folgende Anwendungen:
  • Stimulation von Differenzierungscluster (CD4+)-T-Zellen[1]
  • Aktivierung von primären Lymphozyten des peripheren Bluts[2]
  • Stimulation von mononuklearen Zellen des peripheren Bluts[3][4]

Biochem./physiol. Wirkung

PHA besteht aus zwei Molekularspezies, einem Erythroagglutinin (PHA-E) mit niedriger mitogener und hoher erythroagglutinierender Aktivität sowie einem Leucoagglutinin (PHA-L) mit hoher mitogener und hoher leucoagglutinierender Aktivität, aber sehr niedriger erythroagglutinierender Aktivität.
PHA-E ist nicht blutgruppenspezifisch, die Agglutination kann aber durch bestimmte Oligosaccharide gehemmt werden. PHA-P ist die Proteinform von PHA vor Trennung und Aufreinigung von Erythroagglutinin und Leucoagglutinin. PHA-M ist die Mucoproteinform. Konjugate werden aus den entsprechenden aufgereinigten Lektinen zubereitet.

    1. Generally, PHA-P or PHA-L is utilized to stimulate lymphocytes, although PHA-M will also work, but not to the same degree. The most active form is PHA-L (the mitogenic concentration is < 5μg/ml). PHA, or Phytohemagglutinin, comprises two molecular species:

      1. Erythroagglutinin (PHA-E), which demonstrates low mitogenic activity and high erythroagglutinating activity.

      2. Leucoagglutinin (PHA-L), which possesses high mitogenic and leucoagglutinating activity, accompanied by very low erythroagglutinating activity.

      PHA-P represents the protein form of PHA before the erythroagglutinin and leucoagglutinin components are separated and purified. PHA-M refers to the mucoprotein form of Phytohemagglutinin.

    1. The stock solution of this product can be prepared in phosphate buffered saline at pH 6.8, resulting in a clear to slightly hazy solution at a concentration of 5 mg per mL. This stock solution can then be diluted in cell culture medium to a working concentration of 10 micrograms of lectin per mL for use in peripheral blood cell culture.

    1. After reconstitution in PBS, this product can be stored at 2-8°C for 2-4 weeks. PHA loses approximately 30% activity after 1 month. Stock solutions of at least 1 mg/mL can be stored in frozen aliquots. Do not dilute to the working concentration until ready to use. Vortex stock solutions prior to use.
