A PHA consiste em duas espécies moleculares, uma eritroaglutinina (PHA-E), que tem baixa atividade mitogênica e alta atividade de eritroaglutinina, e leucoaglutinina (PHA-L), que tem alta atividade mitogênica e leucoaglutinante, porém atividade eritroaglutinante muito baixa.
A PHA-E não é específica do grupo sanguíneo, mas a aglutinação pode ser inibida por certos oligossacarídeos. A PHA-P é a forma proteica da PHA antes da separação e purificação da eritroaglutinina e da leucoaglutinina. A PHA-M é a forma de mucoproteína. Os conjugados são preparados a partir das lectinas purificadas correspondentes.

Agglutination activity is expressed in μg/ml and is determined from serial dilutions of a 1 mg/ml solution. This activity is the lowest concentration to agglutinate a suspension of either human erythrocytes (2% in phosphate buffered saline, pH 6.8) or human leukocytes (107 per mL in saline) after 1 hr incubation at 25 °C.

    1. Catalog numbers L1668 and L8902 are suitable for cell culture assays and cell stimulation. The end user needs to design and optimize the assays. These products have not been specifically tested for intracellular cytokine staining assays. It is recommended to review the literature for further information.

  2. What do the designations -P, -L, -M, and -E represent in the names of PHA products?

    1. Generally, PHA-P or PHA-L is utilized to stimulate lymphocytes, although PHA-M will also work, but not to the same degree. The most active form is PHA-L (the mitogenic concentration is < 5μg/ml). PHA, or Phytohemagglutinin, comprises two molecular species:

      1. Erythroagglutinin (PHA-E), which demonstrates low mitogenic activity and high erythroagglutinating activity.

      2. Leucoagglutinin (PHA-L), which possesses high mitogenic and leucoagglutinating activity, accompanied by very low erythroagglutinating activity.

      PHA-P represents the protein form of PHA before the erythroagglutinin and leucoagglutinin components are separated and purified. PHA-M refers to the mucoprotein form of Phytohemagglutinin.
