Descrição geral

Giemsa is a versatile polychromatic stain, which is suitable for staining a diverse range of specimens. It is a buffered thiazine-eosinate solution designed to provide coloration of blood cells. Giemsa’s stain is frequently used for diagnostic purposes in hematology to differentiate the nuclear and cytoplasmic morphology of platelets, RBCs, WBCs, and parasites. It is frequently used in combination with other dye solutions as May-Grunwald’s solution for Pappenheim (MGG) and Wright-Giemsa. The Giemsa stain was adapted to histology due to its unique staining of chromatin, nuclear membranes, and cytoplasmic elements. Giemsa′s staining solution is composed of methylene blue, azure, and eosin.

Aplicação

Giemsa stain has been used for the staining of cells and blood films.[1][2]It is required for the staining of azurophil granules and is an excellent stain for inclusion bodies.[3]Its applications can be seen in hematology for blood and bone marrow specimens, bacteriology, clinical cytology specimens, histological biopsies, and tumor samples.

Outras notas

Nuclei will be varying shades of purple. Cytoplasmic staining will be varying shades of blue to light pink. Fine reddish to lilac granules may be present in cytoplasm of some cell types. Basophils will demonstrate dark blue black granules in the cytoplasm. Eosinophils will demonstrate bright orange granules in the cytoplasm. Red blood cells should be pink to orange

    1. Giemsa stain, as defined by the Biological Stain Commission, consists of a polychrome methylene blue and eosin y mixture. When methylene blue undergoes the polychroming process, various homologs of methylene blue are expected to be present. Azure A, Azure B, Azure C, and other homologs may be present. The percentages of the various homologs can and will vary. This product is not tested to determine the identity or concentration of these homologs.
