Protocolos

Assays that predict passive absorption of orally administered drugs have become increasingly important in the drug discovery process. As previously described by Faller and Kansy such assays provide rapid, low cost and automation friendly methods to measure a compound’s passive permeability.

Guidelines for various bioassays on the MultiScreen system, which makes it possible to carry out an assay from sample incubation to scintillation counting in a 96-well plate.

Questions

  1. Could you please explain the meaning of the terms "donor," "acceptor," "receiver," and "collection" in relation to MAIPN4550 and MAIPN4510 filter plates?

    1 answer
    1. The donor plate is also known as the filter plate, while the acceptor plate can be referred to as the acceptor, receiver, or collection plate. For detailed information on the PAMPA assay, you can refer to the provided link. Additionally, a datasheet is available that reviews some of the donor and acceptor plates. The donor plates include MAIPN4550 and MAIPNTR10. An alternative donor plate option is S2EM004M99, which is a 'MA' classic designed plate. It is important to note that any plate starting with 'MS---' in the catalog number cannot be used.

As for the acceptor plates, several options are available, such as MSSACCEPT0R, MATRNPS50, and MSCPNUV40.

Moreover, tech notes for the automation of PAMPA-Lipid on the Sciclone ALH 3000 workstation and the automation of PAMPA-HDM using a MultiScreen® Filter Plate on a Beckman Biomek® FX Workstation are also available. An article titled "Evaluation of the reproducibility of Parallel Artificial Membrane Permeation Assays (PAMPA)" provides further insights into the topic.
