Descrição geral

Microtubules are involved in a wide variety of cellular activities ranging from mitosis and transport events to cell movement and the maintenance of cell shape. Tubulin itself is a globular protein which consists of two polypeptides (alpha and beta tubulin). Alpha and beta tubulin dimers are assembled to 13 protofilaments that form a microtubule of 22 nm diameter. Tyrosine ligase ads a C-terminal tyrosin to monomeric alpha tubulin. Assembled microtubules can again be detyrosinated by a cytoskeleton associated carboxypeptidase. Detyrosinated alpha tubulin is referred to as Glu-tubulin. Another post-translational modification of detyrosinated alpha tubulin is C-terminal polyglutamylation which is characteristic for microtubules in neuronal cells and the mitotic spindle.

Especificidade

Expected to cross-react with mammals, sea urchin, plants, not yeast (Paturle-Lafanechere, L., 1991).
The antibody specifically recognizes the detyrosinated form (Glu tubulin) of the tubulin alpha chain (Bianchi, M., 2006; Goulimari, P., 2005).

Imunogênio

Peptide corresponding to the C-terminal of Glu tubulin.

Aplicação

Anti-Tubulin Antibody, Detyrosinated is an antibody against Tubulin for use in IC, IH(P) and WB.
Immunohistochemistry:
A previous lot was used on either frozen or paraffin embedded, fixed tissue (Gundersen, G.G., 1984): 1:200-1:500 dilution.

Optimal working dilutions must be determined by the end user.

Qualidade

Routinely evaluated by Western Blot on L6 lysate lysates.

Western Blot Analysis: 1:500 dilution of this lot detected detyrosinated tubulin on 10 μg of L6 lysate.

Descrição-alvo

50 kDa

forma física

Affinity purified immunoglobulin
Purified rabbit polyclonal in buffer containing liquid PBS.

Armazenamento e estabilidade

Stable for 6 months at -20°C in undiluted aliquots from date of receipt.

Handling Recommendations:
Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Nota de análise

Control
Hela Whole Cell Lysate.

Cilia have a unique diffusion barrier ("gate") within their proximal region, termed transition zone (TZ), that compartmentalises signalling proteins within the organelle. The TZ is known to harbour two functional modules/complexes (Meckel syndrome [MKS] and Nephronophthisis [NPHP]) defined by genetic

In neurons, the polarized distribution of vesicles and other cellular materials is established through molecular motors that steer selective transport between axons and dendrites. It is currently unclear whether interactions between kinesin motors and microtubule-binding proteins can steer polarized transport.

RAS signalling through phosphoinositide 3-kinase (PI3-Kinase) has been shown to have an essential role in tumour initiation and maintenance. RAS also regulates cell motility and tumour invasiveness, but the role of direct RAS binding to PI3-Kinase in this remains uncertain.
