Beschreibung

Application

Diisopropylfluorphosphat wird wie folgt verwendet:

• zusammen mit inaktiviertem Thrombin in Affi-Gel-10-Säulen zur Aufreinigung von rekombinantem humanem Thrombomodulin mit dessen Chondroitinsulfatkette (shrTMCSA)[1]
• zur Auslösung von Status epilepticus (SE) in adulten Ratten[2]
• als irreversibler Serinproteaseinhibitor in humanen neutrophilen subzellulären Fraktionen[3]

Biochem/physiol Actions

Wirksamer Inhibitor von Serinproteasen und Acetylcholinesterase; hemmt Cathepsin G, Cholinesterase, Gerinnungsfaktor Xa, Leukozyten-Elastase, Pankreas-Elastase, Gewebekallikrein, Plasmin, Subtilisin und Thrombin. Hemmt die durch Rizin und bakterielle Toxine ausgelöste Apoptose.

Wirksamer Inhibitor von Serinproteasen wie Trypsin und Chymotrypsin sowie von Acetylcholinesterase; hemmt außerdem Cathepsin G, Cholinesterase, Gerinnungsfaktor Xa, Leukozyten-Elastase, Pankreas-Elastase, Gewebekallikrein, Plasmin, Subtilisin und Thrombin. Inhibition von Acetylcholinesterase macht diesen Wirkstoff besonders toxisch. Hemmt die durch Rizin und bakterielle Toxine ausgelöste Apoptose.

Analysis Note

Wird normalerweise in einer Konzentration von 0,10 mM verwendet. Phenylmethylsulfonylfluorid (PMSF) ist eine sicherere Alternative zur Inhibition von Serinproteasen.

Other Notes

DIFP ist bei ordnungsgemäßer Lagerung in einer ungeöffneten Flasche bei 2–8 °C für mindestens zwei Jahre haltbar. DIFP
ist instabil, wenn es Feuchtigkeit ausgesetzt wird. DIFP nimmt bei Zersetzung eine tiefgelbe Farbe an.
