Beschreibung

General description

Eine zellpermeable Chinolyloxyphenyl-Harnstoff-Verbindung, die sowohl in zellbasierten (IC₅₀ = 0,9 nM bei VEGF-induzierter Rezeptorphosphorylierung) als auch in zellfreien (IC₅₀ = 4,0 nM bei Kinasereaktionen) Assays als VEGFR-2-selektiver Inhibitor wirkt. Sie hemmt c-Kit, PDGFRα und FGFR-2 nur bei sehr viel höheren Konzentrationen (IC₅₀ = 40, 67 bzw. 170 nM in zellbasierten Assays), während sie gegen acht andere häufig untersuchte Rezeptor- und Nicht-Rezeptor-Kinasen selbst bei Konzentrationen von 10 µM keine Wirkung zeigt. Sie blockiert wirksam die VEGF-abhängige (aber nicht die VEGF-unabhängige) Zellproliferation in vitro und das Tumorwachstum bei Mäusen und Ratten (5 bis 20 mg/kg, täglich p. o.) in vivo.
