Beschreibung

General description

Eine zellpermeable Diarylthiazol-Verbindung, die die Zellaktivierung der SREBP-1/2-Vorläufer (Sterol Regulatory Element Binding Proteins 1/2) in die aktiven Zellkernformen hemmt (>;95 % Hemmung bei 20 µ;M in CHO-K1-Zellen), indem SREBPs-SCAP (SREBP Cleavage-Activating Protein) ER zu Golgi-Translokationen blockiert werden; geringe Wirkung gegen die Transkriptionsaktivität reifer SREBPs. Aus Bindungsstudien mit rekombinanten SCAP-Restkonstrukten ging hervor, dass Fatostatin SCAP zwischen aa 448 und 767 reversibel anzielt und keine Wirkung auf die SREBP-SCAP-Interaktion hat. Fatostatin unterdrückt effektiv die SREBPs-abhängigen Zellfunktionen in vitro, darunter insulin-induzierte 3T3-L1-Adipogenese (vollständige Hemmung bei ≤;20 µ;g/ml) und IGF1-abhängige DU-145-Vermehrung (IC₅₀ = 100 nM) und weist in vivo eine Wirkung auf den Blutzuckerspiegel (Senkung um 70 %) und das Körpergewicht (Senkung um 12 %) von ob/ob-Mäusen auf, die über 28 Tage hinweg (30 mg/kg täglich intraperitoneal), ohne Auswirkungen auf die Nahrungsaufnahme der Tiere oder anscheinende unerwünschte Wirkung.
