Beschreibung

General description

Eine zellpermeable Pyrazolopyrimidinylamino-Verbindung, die als potenter, ATP-kompetitiver und reversibler Inhibitor von CKIε dient, mit einer ~22-mal größeren Potenz als für CKIδ (IC₅₀ = 32 bzw. 711 nmol/l) und eine ausgezeichnete Selektivität unter 50 Kinasen aufweist. Die einzige Ausnahme ist EGFR (69%ige Hemmung bei 1 µmol/l). Blockiert die Zellkerntranslokation von PER3 (PERIOD3) in COS7-GFP-PER3-CKIε-Zellen im Gegensatz zu mit CKIδ transfizierten Zellen (IC₅₀ = 0,13 bzw. 2,65 µmol/l) und den PER2-Abbau in COS7-GFP-PER2-CKIε-Zellen bevorzugt. Anders als Casein-Kinase I-Inhibitor II, PF-670462 (Katalognr. 218716), übt PF-4800567 unter ähnlichen Bedingungen nur einen geringen Einfluss auf die innere Uhr aus.
