General description

RIPA (Radio-Immunoprecipitation Assay) Buffer is supplied as a ready to use solution that requires no preparation. It minimizes non-specific protein-binding interactions to keep background low, while allowing most specific interactions to occur, enabling studies of relevant protein-protein interactions. It is widely used in applications such as immunoprecipitation since most antibodies and protein antigens are not adversely affected by the components of this buffer.
Radioimmunoprecipitation assay buffer (RIPA buffer) is a lysis buffer used for rapid, efficient cell lysis and solubilization of proteins from both adherent and suspension cultured mammalian cells.

Application

RIPA Buffer enables efficient cell lysis and protein solubilization while avoiding protein degradation and interference with the proteins′ immunoreactivity and biological activity. RIPA Buffer also results in low background in immunoprecipitation and molecular pull-down assays. Compatible with EZview™ Affinity Gels.
RIPA Buffer has been used as a reagent for cell lysis in the following experimental studies:
  • Fibroblast-based deep phenotyping of the peripheral tissue, which facilitates the mechanistic disease stratification in sporadic Parkinson′s disease (sPD).[1]
  • Demonstrating the inhibition of oxidative phosphorylation (OXPHOS), which can activate the mixed lineage kinase domain-like protein (MLKL)-dependent necroptosis in human lung epithelial cells.

Features and Benefits

1 mL of the RIPA Buffer is used to lyse cells from one 100 mm culture dish (0.5 to 5 * 107 cells) of most adherent mammalian cell lines.
Minimizes non-specific protein binding interactions to ensure low background.

Other Notes

Ready-to-use solution containing 150 mM NaCl, 1.0% IGEPAL® CA-630, 0.5% sodium deoxycholate, 0.1% SDS, 50 mM Tris, pH 8.0.

  1. we bought a bottle of RIPA buffer and want to make alliquots of this bottle. Can we store the alliquots of the buffer by -20? Or is it better to keep them between 2°C-8°C ?

    1. It is recommended to store the product at 2–8C. Studies have not been performed on aliquots and to determine if refrigerated storage will extend the shelf life of this product. We would recommend searching the general research literature or contacting researchers who routinely use this product to see if solution stability information is available. If a recommended storage is not given in the literature, it is usually best to make solutions fresh for use. If you decide to store the aliquots at -20°C, we would recommend validating activity of the supplemented buffer after freezing and avoid any thawing/freezing cycle.   5. How long can the RIPA buffer be stored in 4C? I stored it for almost 6 years, can I still use it?

    1. Regarding stability in the recommended temperature of 2-8 °C, this product is not assigned an expiration date or recommended retest date. Products with no expiration date or recommended retest date should be routinely inspected by customers to ensure they perform as expected. These products are also subject to a one year warranty from the date of shipment.     1. R0278 can be used on tissue, if it is well homogenized.

  8. Does Product R0278, RIPA buffer, contain any protease inhibitors or phosphatase inhibitors?

    1. This buffer does not contain any protease or phosphatase inhibitors, however, they may be added to insure against possible proteolysis.

  9. I left the bottle of Product R0278, RIPA buffer, on my bench for about a week. Can I still use it?

    1. Yes, you should be able to use this product.  Although we recommend storage in the refrigerator at 2-8°C for long-term storage, Product No. R0278 may be kept at ambient temperature (our normal shipping condition) for a short time (approximately 2 weeks).
