转铁蛋白主要在肝脏中合成,是血清中含量丰富的糖蛋白。转铁蛋白超家族成员具有相似的多肽折叠模式。转铁蛋白含有N端和C端铁结合同源结构域。这些结构域中的每一个都被分裂成两个子结构域,在子结构域间隙内具有铁和阴离子的结合位点。绑定间隙随着铁释放和铁绑定打开和关闭。脱铁转铁蛋白是一种无铁蛋白,在转铁蛋白与其受体分离后产生。此产物可以补充铁或用于结合培养基中存在的游离铁。

應用

脱铁转铁蛋白已用于:
  • 培养人原代胰腺内皮细胞、Het1As(非肿瘤食管细胞)和永生化人结肠上皮细胞(HCEC-1CT)
  • 培养α小鼠肝12(AML-12)(成熟肝细胞)细胞
  • 作为晶体学前纯化的人脱铁转铁蛋白的来源

生化/生理作用

转铁蛋白负责将铁从吸收和储存的部位运送到组织细胞。它能维持生物体液中的铁含量。转铁蛋白的水平可以反映总铁结合能力(TIBC)。转铁蛋白为骨髓中红细胞的血红蛋白提供所需的铁元素。缺铁会导致转铁蛋白水平升高。怀孕和服用雌激素也会提高转铁蛋白的水平。慢性肝病、肾功能不全、营养不良和蛋白质丢失性肠病会减少转铁蛋白的合成。
