植物总蛋白抽提试剂盒(Plant Total Protein Extraction Kit)是专为植物生命科学应用而设计的产品,能够从任意植物种类或组织中抽提蛋白定性样本。本方案无需任何超速离心步骤或含水多聚物的两相分离步骤(APTP)。本试剂盒包含两类试剂:一份植物特异性的蛋白酶抑制剂cocktail、一份增加了溶解能力以抽提更多疏水蛋白质的全新离液剂(C0356, 蛋白抽提试剂第4类试剂,Protein Extraction Reagent Type 4). 在植物组织样品顺序蛋白抽提过程中采用新鲜试剂,能洗脱植物组织中的几乎所有蛋白质。蛋白酶抑制剂cocktail是作用于丝氨酸、半胱氨酸、金属蛋白酶、天冬氨酸与氨肽酶等酶类的多种蛋白酶的广谱抑制剂混合物。本品含有胃酶抑素A,4-(2-氨基乙基)苯磺酰氟(AEBSF)中,E-64,亮肽素,1,10-菲咯啉,和苯丁抑制素等多种成分。经验证,cocktail抑制剂混合物能够在抽提过程中高效避免蛋白降解的发生。

在去除了多酚,鞣质等干扰物质之后,新鲜或冻存的植物组织经研磨后,用离液剂重悬。通过离心沉淀植物碎片,并收集蛋白抽提液。最后用户可获得定性的分级蛋白样品,用于下游蛋白组学分析。
