一般說明
胰蛋白酶(EC 3.4.21.4)是一种胰腺丝氨酸蛋白酶,能够特异性剪切精氨酸和赖氨酸残基羧基侧的肽键。如果剪切位点的任意一侧存在酸性残基,则水解速率降低;如果羧基侧存在脯氨酸残基,则可能不会发生剪切。对目标蛋白质进行胰蛋白酶消化,可产生高度特异性的剪切以及数量有限的肽段。
適合性
适用于检测各种样品中的胰蛋白酶活性。
原則
在该检测中,胰蛋白酶可切割底物以产生 p-硝基苯胺(p-NA),其可在λ= 405nm处进行检测。由于颜色强度与p-NA含量成正比,因此可以对胰蛋白酶活性进行准确测定。该试剂盒可在各种样品中检测10-100mU(p-NA单位)胰蛋白酶。
