肾素，也称为血管紧张素原酶，是一种天冬氨酰蛋白酶，可切割血管紧张素原，形成血管紧张素 I，这是肾素-血管紧张素系统（RAS）的初始和限速步骤。该系统在血压和体液平衡的调节中起关键作用。由于血管收缩和液体潴留增加，肾素活性增加可导致高血压。肾素抑制剂可用于高血压的治疗。[1]

适用性

肾素检测试剂盒提供了一种方便、简单的检测方法，适于筛查多种样品中的肾素活性，或用于筛选肾素抑制剂。

原理

肾素的活性是通过含有荧光猝灭剂的专有荧光共振能量转移（FRET）肽测定的。肾素切割肽会产生一种荧光产物（λex = 540/λem = 590 nm）。该测定的灵敏度约是基于 EDANS/DABCYL 的测定的 50 倍。

    1. Although the standard that has been tested is recombinant human renin, theoretically the kit should be suitable for all other species.

  2. Does the kit MAK157 contain any Renin inhibitor? If so, how much Renin inhibitor should be added, and at what step of the assay should it be added in order to determine the Renin activity?

    1. The kit MAK157 does not contain any Renin inhibitor. To confirm the activity using this kit, researchers can use a RENIN INHIBITOR separately.The amount of inhibitor to use and the step to use the Renin inhibitor can be referred to from published literature.

    Kit instructions misleading - be careful

    Kit provides you with 10mL assay buffer. At least 1.87mL needed for making standards. 5mL needed for making substrate. Leaves you with 3.13mL for diluting samples. If your samples need a lot of diluting this is insufficient. I did not find the customer service suggestion of using water to dilute my samples to be appropriate or helpful. They could instead just provide sufficient reagent. God help you if you want to run the assay twice, with two sets of standards, further depleting the assay buffer stock.
