谷氨酸脱羧酶（GAD; E.C.4.1.1.15）是负责将谷氨酸转化为γ-氨基丁酸（GABA）的一种酶，γ-氨基丁酸是大脑较高区域的主要抑制递质，在胰岛中被认为是一种旁分泌激素。GAD的两种分子形式（65 kDa和67 kDa，两种形式之间的氨基酸相似度为64％）是高度保守的，两种形式都在CNS、胰岛细胞、睾丸、输卵管和卵巢中表达。两种亚型在大鼠和小鼠的脑细胞质中分布（Sheikh，1999）。GAD65是一种两亲性膜锚定蛋白（585个氨基酸），由人类10号染色体进行编码，负责囊泡GABA的产生。GAD67位于细胞质中（594个氨基酸），由2号染色体进行编码，其可能是导致大量细胞质GABA产生的原因。GAD的表达在大鼠脊髓神经发育过程中发生变化。GAD65在E13周围的连合轴突中瞬时表达，但在第二天被下调，而GAD67的表达主要在这些神经元的躯干中发生上调（Phelps，1999）。在成熟的大鼠胰腺中，GAD65和GAD67似乎存在差异化定位，GAD65主要存在于含胰岛素的β细胞中，而GAD67存在于含胰高血糖素的（A）细胞中（Li，1995）。GAD67的表达具有特别的可塑性，并且可以响应实验操作（例如神经元刺激或横切）或疾病进展以及诸如精神分裂症等紧急疾病而发生变化（Volk，2000）。两种GAD亚型的共定位也显示出GAD65/GAD67分布的变化，这一变化与某些疾病状态（例如IDDM和SMS）相关。

特异性

抗GAD67抗体，克隆1G10.2可与大鼠、小鼠和人类来源的谷氨酸脱羧酶（GAD67）的67kDa亚型发生反应，其他物种尚未进行测试。与使用AB1511（其可与GAD65和GAD67发生反应）进行印迹检测相比，在大鼠脑裂解液上通过蛋白质印迹该抗体未检测到与GAD65的交叉反应性。

在4％低聚甲醛固定的小鼠和大鼠组织上的免疫组织化学分析：1:1,000-1:5,000.该抗体以1：2,500-1：5,000的稀释度也可用于人类组织（4％PFA，4°C或100％乙醇，+20°C或100％丙酮，-20°C，固定十分钟）。
GAD67的多克隆产品也以AB5992编号进行销售。

最佳工作稀释度必须由最终用户进行确定。
该抗GAD67抗体，克隆1G10.2，已经过验证可用于IH、IH（P）和WB中对GAD67进行检测。该抗GAD抗体已经有超过75次产品引用。

质量

已通过SKNSH细胞裂解液的免疫组织化学进行常规评估。

免疫组织化学（石蜡）分析：
GAD67在小鼠大脑皮层体感1区的代表性染色模式和形态。 所有的褐色斑点均为层状VI神经元。无需检索表位。 使用IHC Select时，将该批次抗体稀释至1：500 使用HRP-DAB进行检测。
