台盼蓝(C.l.23850)是一种大的四磺化阴离子亲水染料。它也被称为二胺蓝 3B。

應用

台盼蓝已被用作台盼蓝排斥试验/细胞活力试验中的染料,以检测死细胞。[1][2]

生化/生理作用

台盼蓝被大多数活细胞阻挡,但可被带入吞噬细胞和某些其他细胞。

该染料可根据′细胞吸收或排除染料的能力来区分死细胞和活细胞。活细胞应具有完整的细胞膜,因此将这种染料排除在外,并且细胞质是透明的。另一方面,没有活力的细胞显示出蓝色的细胞质。
台盼蓝通常用于各种细胞类型的活性测试,包括冷冻精子和接触杀真菌药物的主动脉肌细胞等。它也在Van Gieson手术中用作胶原蛋白染色。对于淀粉样蛋白染色,台盼蓝被认为是Puchtler-Bennhold染色中刚果红的蓝色替代品。 它适用于细胞活力测定:
  • 通过用 0.2% 台盼蓝溶液染色来确定活细胞和坏死细胞的数量。

  1. Are the descriptions of Product 302643 as "Trypan Blue - 60% Dye content" and the purity limit of ">= 80.0%" on the Specification sheet and CofA referring to the same thing? If they are, why are they different? If not, should both be considered when aiming to achieve a specific dye concentration? Additionally, if it is 60% dye, what makes up the remaining 40%? Should the significant water content in this product be taken into consideration?

    1. Dye content is determined by back calculation from the carbon or nitrogen content, while purity is established by HPLC. The dye content is 60%. The impurities are not identified by either dye content or by methods such as HPLC, and no testing for impurities is conducted in this case. Both dye content and dye purity are determined for this product, with the Specification sheet listing minimum specifications for all lots, and individual Certificates of Analysis reporting lot-specific results. Water content is not measured, and the product is not considered hygroscopic.
