There is alpha Naphthol kit in hand and the procedure 91, it looks like there should be more components in the kit. Interested in the other methods listed in the procedure sheet but only have reagents for the ANAE staining. Please clarify.

How can human kidney tissue be stained with the PAS stain using product TTR009? It appears to be embedded and did not take on the stain. The intended application is for the cytochemical staining of lizard blood cells.

The PAS Tissue Trol slides need to have the paraffin removed before staining. In general, it is best to match the specimen to the control. Since the PAS Tissue Trol slides have been embedded in paraffin, the paraffin will need to be removed first. The recommended control tissue for staining would be a human peripheral blood smear. Neutrophils typically stain moderately strong with PAS, while other cell types may show varying degrees of staining or none at all. The recommendation for human blood as a control is based on the expectation that human neutrophils should make a good control, and it is anticipated that this may be true for lizards as well. However, since there is no experience with lizards, human blood is recommended. Different species may have varying amounts of a substance in their blood, leading to variations in staining. Therefore, it may be necessary to modify the procedure to achieve similar staining.
