General description

Giemsa is a versatile polychromatic stain, which is suitable for staining a diverse range of specimens. It is a buffered thiazine-eosinate solution designed to provide coloration of blood cells. Giemsa’s stain is frequently used for diagnostic purposes in hematology to differentiate the nuclear and cytoplasmic morphology of platelets, RBCs, WBCs, and parasites. It is frequently used in combination with other dye solutions as May-Grunwald’s solution for Pappenheim (MGG) and Wright-Giemsa. The Giemsa stain was adapted to histology due to its unique staining of chromatin, nuclear membranes, and cytoplasmic elements. Giemsa′s staining solution is composed of methylene blue, azure, and eosin.

Application

Giemsa stain has been used for the staining of cells and blood films.It is required for the staining of azurophil granules and is an excellent stain for inclusion bodies.Its applications can be seen in hematology for blood and bone marrow specimens, bacteriology, clinical cytology specimens, histological biopsies, and tumor samples.
Certified for Giemsa stain for blood films and for the Wolbach′s Giemsa method for paraffin sections.

Other Notes

Nuclei will be varying shades of purple. Cytoplasmic staining will be varying shades of blue to light pink. Fine reddish to lilac granules may be present in cytoplasm of some cell types. Basophils will demonstrate dark blue black granules in the cytoplasm. Eosinophils will demonstrate bright orange granules in the cytoplasm. Red blood cells should be pink to orange

  1. Hello, is this product suitable for G banding of human chromosomes? Thank you!

    1. G5637 is sent to the Biological Stain Commission (BSC) for certification as a biological dye. The BSC performs various tests and also tests suitability for use in staining blood films and in the Wolbach's Giemsa method for paraffin embedded tissue. There is no performance testing for the G Banding of human chromosomes. The customer would need to provide their own staining protocol or locate a published method if they intend to use G5637 for the G banding of human chromosomes.

  2. Hello, could you confirm the dye(s) present in G5367? Is it only the pictured compound (I think Azure A) without methylene blue or eosin? Thank you!

    1. Giemsa stain, as defined by the Biological Stain Commission, consists of a polychrome methylene blue and eosin y mixture. When methylene blue undergoes the polychroming process, various homologs of methylene blue are expected to be present. Azure A, Azure B, Azure C, and other homologs may be present. The percentages of the various homologs can and will vary. This product is not tested to determine the identity or concentration of these homologs.
