Although other uses of P8920 have been reported, there is no data on hand regarding the use of P8920 to make the DNA binding stronger on polystyrene MaxiSorp plates.

Aplicación
La disolución de poli-L-lisina se ha utilizado para la fijación de cortes transversales de células endoteliales a membranas de PET en marcos de acero reticulados mediante UV.[1] Los marcos recubiertos de poli-L-lisina se utilizaron para la colocación de cortes de tejido para análisis inmunohistoquímicos.[2]
Los polímeros de poli-L-lisina pueden utilizarse en la promoción de la adhesión celular a los sustratos sólidos, la conjugación al metotrexato para incrementar el transporte de medicamentos, la microencapsulación de islote, la tecnología de microencapsulación celular, recubrimiento de portaobjetos de vidrio de micromatrices y preparaciones cromosómicas. La poli-L-lisina de menor peso molecular (30 000-70 000) es menos viscosa en disolución, pero las versiones de mayor peso molecular proporcionan mas sitios de unión por molécula.
Acciones bioquímicas o fisiológicas
La poli-L-lisina es un factor de unión inespecífica a las células útil en la promoción de la adhesión celular a los sustratos sólidos intensificando la interacción electrostática entre iones con carga negativa de la membrana celular y la superficie del cultivo. Cuando se absorbe a la superficie del cultivo celular, la poli-L-lisina funciona aumentando el número de sitios con carga positiva disponibles para la unión a la célula. Con las células que pueden digerir la poli-L-lisina, debe utilizarse la poli-D-lisina como factor de fijación.
Componentes
La poli-L-lisina es una polímero de aminoácido con carga positiva con aproximadamente un HBr por residuo de lisina. El hidrobromuro permite a la poli-L-lisina estar en una forma cristalina hidrosoluble. Puede encontrarse una pequeña cantidad del producto en la estructura beta porque el HBr interfiere en el enlace de hidrógeno entre el grupo amino y los grupos carboxilo o los grupos que contienen N u O.
Nota de preparación
Este producto se suministra como una disolución al 0,10 % (p/v) en agua. El recubrimiento de los portaobjetos con esta disolución debe ir seguido de una incubación de 5 minutos y de secar los portaobjetos a temperatura ambiente. Los portaobjetos recubiertos serán estables durante un año si se protegen del polvo.

Solutions of this product are not sterile and contain a preservative, ProClin, to control contamination. In theory you could sterile filter P8920 through low protein binding filters, such as PVDF or PES. Sterile filtering will not remove the ProClin from P8920. Live cells should not be exposed to ProClin, as ProClin interrupts the Krebs cycle at 4 different sites. It would be best to purchase the powder as P1524 and then sterile filter after the solution has been prepared.

The number of slides that may be coated is dependent upon the surface area. The general use instructions recommend using 0.5 - 1.0 mL per 25 cm2 coating area - the 25 cm2 value is based on a T-25 tissue culture flask. A standard microscope slide has an approximate area of 1.3 cm2.

Is Product P8920, Poly-L-lysine solution, recommended for cell culture use?

No, the product contain 0.01% thimerosal as a preservative. For cell culture use, product P4832 is recommended. This solution has the same molecular weight as P8920 but contains no preservative. P4832 is sterile filtered and is considered suitable for cell culture use.

What are the recommended uses for Product P8920, Poly-L-lysine solution?

P8920 was developed for use in coating slides intended for use in either histology or immunohistochemistry. Poly-L-lysine is preferred for uses such as silver staining procedures that require an alkaline pH. Silane coated slides may be more effective is a low pH is required.
