一般說明

吐温20是聚氧乙烯山梨醇酯,属于聚山梨醇家族。这是一种分子量为1225道尔顿的非离子洗涤剂,认为其含20个环氧乙烷单位、1个山梨醇和1个月桂酸为主要脂肪酸。环氧乙烷亚基提供了表面活性剂的亲水性,而碳氢化合物链提供疏水环境。山梨醇形成连接环氧乙烷聚合物的骨架环。

應用

吐温20已可用于 原位 杂交的染色缓冲液中。在荧光共振共振能量转移(FRET)实验样品制备过程中,它也被用来防止蛋白质对盖玻片的非特异性吸附。
吐温20是一种广泛应用于生化领域的非离子去污剂。它被用作稳定的水包油乳液的乳化剂。吐温20已可用于膜的预提取,以去除周围蛋白(用于提取膜结合蛋白的比例为2%)。
吐温20在基于膜的免疫分析中已被用作一种封闭剂,其常用浓度为0.05%。吐温20可用于裂解哺乳动物细胞,使用浓度为0.005至0.5%。
非离子洗涤剂

注意

吐温20在高压灭菌下可能不稳定,特别是在含金属阳离子的缓冲溶液中。如果没有对性能的变化进行测试,则不建议使用高压灭菌。吐温20对热敏感,放置于升高的温度中其颜色会变深。据报道,聚山梨醇酯与碱、重金属盐、酚和单宁酸不相容。它们可能会降低许多防腐剂的活性。聚山梨醇酯水溶液在贮存过程中发生自氧化反应,光、温度升高和硫酸铜催化其变化。

準備報告

吐温20可混溶于水(100mg/mL),得到透明的黄色溶液。它也可以与酒精,二恶烷,乙酸乙酯混溶。吐温20不溶于液体石蜡和固定油脂。
