一般說明

乳酸脱氢酶（LDH）的总分子量为140kDa，由4个亚基组成，称为M亚基（肌肉）和H亚基（心脏）。这些亚基可以以5种组合（M4、M3H1、M2H2、MH3和H4）中的任何一种进行混合。 骨骼肌含有LDH，其主要是M4，含有少量M3H和微量的H2H2。 H和M亚基的分子量非常相似，但氨基酸组成十分不同。兔肌肉LDH在pH5.0的乙酸盐-氯化物中可解离为二聚体物质（MW = ~70kDa），该解离是可逆的。Biochemistry, 13, 3527-3531 (1974). 氧化乙醛酸和乳酸。

應用

来自兔子肌肉的L-乳酸脱氢酶已用于：
  • 蛋白结合测量[1]
  • 检测草酰乙酸脱羧酶的作用[2]
  • 确定血清中的 L-和 D-乳酸[3]

生化／生理作用

乳酸脱氢酶负责在发酵代谢中将丙酮酸转化为乳酸。[4]
其还可催化其他L-2-羟基单羧酸的氧化。

單位定義

在pH7.5，37℃条件下，一个单位每分钟可将1.0μmole的丙酮酸还原为 L-乳酸。

外觀

溶于3.2 M（NH4）2SO4溶液中的结晶悬浮液，pH 6.0
