一般描述

弹性蛋白酶是一种蛋白水解酶,属于肽基肽水解酶亚组成员。它是动脉的主要解剖组分。[1]主要存在于各种鸟类和哺乳动物的胰腺和胰液中,还存在于人血清、粒细胞和红细胞中。[2]

应用

弹性蛋白酶(来源于人白细胞)已用于:
  • 测定血清弹性蛋白酶活性[3]
  • 纤连蛋白和唾液腺的蛋白水解消化[4]
  • 中性粒细胞弹性蛋白酶(NE)活性测定[5]

弹性蛋白酶已用于消化纤连蛋白。将所得消化片段与人白细胞粗制匀浆的纤连蛋白消化片段进行比较,可用于研究干燥综合征患者唾液中纤连蛋白肽的存在。[4]它还用作细胞裂解物的弹性蛋白酶活性测定的对照,用于研究全反式视黄酸对培养的原始细胞的促凝血和纤溶活性的影响,该原始细胞来自急性早幼粒细胞白血病患者。[6]

生化/生理作用

弹性蛋白酶在蛋白水解过程的辅助下可从不溶性弹性蛋白纤维中释放可溶性多肽。[7]它在腺苷三磷酸(ATP)的辅助下可激活轴丝解体。[8]与胰腺弹性蛋白酶不同,白细胞酶优先切割缬氨酸的羧基侧,一定几率下也会在丙氨酸基团后进行切割。天然底物包括弹性蛋白、软骨蛋白聚糖、I、II、II和IV型胶原以及纤连蛋白。

物理属性

白细胞弹性蛋白酶是蛋白酶 S1 家族的一个 29 kDa 的丝氨酸内切蛋白酶。以单个 238 个氨基酸-肽链形式存在,有 4 个二硫键。其含有两种或多种组成的 N-连接聚糖,这解释了其三种主要亚型。
