生物化学的／生理学的作用
p53は、形質転換の阻害能や腫瘍細胞増殖の抑制能を示し、腫瘍抑制因子として確認されています。さらにp53は、下流遺伝子のサブセットの発現を調節できる、転写因子でもあります。p53中の2つの特定のN末端残基(Leu22、Trp23)が突然変異すると、p53によるトランス活性化が低下します。さらに、TAFII40とTAFII60(またはTAFII31とTAFII70)への結合能とも関係していることは(7、8)、これらの相互作用の1つ、または両方が、活性化に重要であることを示唆しています。22および23残基がAlaに突然変異しても、TBPへの結合能には影響しません。しかしこれらの残基がアミノ酸に変異したとき、p53-TBP相互作用が失われたことが報告されています。p53遺伝子の様々な突然変異は、多様ながん種において特徴づけられています。p53機能の低下または変異は、腫瘍形成と高度に相関しています。
