C-X-C motif chemokine ligand 12 (CXCL12), also known as stromal cell-derived factor 1 α (SDF-1a), is encoded by the gene mapped to human chromosome 10q11.[1] Stromal cell-derived factor 1 α and β (SDF-1 α and β) are stromal derived CXC chemokines and signal through the C-X-C motif chemokine receptor-4 (CXCR4).[2] CXCL12 is mainly expressed in the inflamed and failing myocardium.[3]

アプリケーション

Stromal Cell-Derived Factor 1β has been used to activate chemokine receptor-4 (CXCR4) and to further assess its role in tumor cell migration using chemotaxis assay.[4]

生物化学的／生理学的作用

SDF-1/PBSFは、α(CXC)ケモカインファミリ-の分子であり、Tリンパ球および単球の走化性因子として作用しますが、好中球に対する効果はありません。SDF-1αとSDF-1β(SDF-1/PBSFとしても知られています)は1本の遺伝子によりコ-ドされており、オルタナティブスプライシングにより生じます。これら2種類のタンパク質は、SDF-1βのC末端に存在する4個のアミノ酸残基がSDF-1αには存在しないという点を除いて一致しています。SDF-1/PBSFは種間で高度に保存されており、ヒトとマウスのタンパク質の場合、アミノ酸の違いはわずか1残基です。

C-X-C motif chemokine ligand 12 (CXCL12) is involved in inflammation, proliferation, tumorigenicity and metastasis.[5] CXCL12 exerts HIV (human immunodeficiency virus) suppressive activity in cells expressing the CXCR4 (C-X-C motif chemokine receptor 4).[6] It is overexpressed in breast carcinoma, ovarian carcinoma and papillary thyroid carcinoma. It enhances angiogenesis in the ischemic tissue. SDF1 is also implicated in neuroprotection after ischemic stroke with the help of CXCR4.[7][8]
