General description

アビジンは、トリ、爬虫類、および両生類の卵管で産生される4量体または2量体のビオチン結合タンパク質であり、これらの卵白中に蓄積されます[1]。ビオチンに対する4つの高親和性結合部位で構成されます。ExtrAvidin®は、卵白アビジンから作成されます。アビジンからアフィニティ精製された、アビジンの高い親和性とストレプトアビジンの低バックグラウンド性を持った精製アビジンです。細菌Streptomyces avidinii.によって産生されるビオチン結合タンパク質です[2]。アルカリホスファターゼと結合させたExtrAvidinは、様々なテクニックに使用されます。

Application

ExtrAvidin® -アルカリホスファターゼは、以下に使用されています：
  • ドットブロット
  • 免疫組織化学（ホルマリン固定、パラフィン包埋切片）
  • 間接的ELISA
  • 酵素結合免疫吸着測定法（ELISA）[3][4]
  • 酵素結合免疫吸着スポット（ELISPOT）測定法[5]
  • 競合的ELISA[6]

Biochem/physiol Actions

アルカリホスファターゼは、エステルからリン酸への加水分解を触媒します。また、膜を通しての基質の輸送を促進します。

Physical form

0.05M Tris-HClバッファ-溶液 (pH 8.0, 1mM MgCl2, 1%BSA, 15mM アジ化ナトリウム含有)。
