ターゲットの説明
シトクロムcは、アポト-シス初期の関連事象としてミトコンドリアから放出される電子伝達タンパク質です。シトクロムcおよびdATPは、Apaf-1、Bcl-2、プロカスパ-ゼ9で構成される哺乳類アポトソ-ムのコファクタ-です。プロカスパ-ゼ9の活性化とともに、デスプロテア-ゼであるカスパ-ゼ3を含めた他のカスパ-ゼが活性化されていきます。Bcl-2の存在によって細胞小器官におけるシトクロムcの放出が阻害され、アポト-シスの開始が阻止されます。UV照射、スタウロスポリン、Bax過剰発現などさまざまな方法でアポト-シス誘導された細胞において、カスパ-ゼインヒビタ-によるシトクロムc放出の抑制効果はみとめられていません。
