Descrizione generale

The complement system consists of many plasma proteins that assist the ability of phagocytic cells and antibodies to clear pathogens. There are three different simultaneous pathways. The classical pathway is stimulated by antigen-antibody complexes; the alternative pathway spontaneously activates on contact with pathogenic cell surfaces; and the mannose-binding lectin (MBL) pathway recognizes mannose sugars usually present only on pathogenic cell surfaces. Because of the potential to be extremely damaging to host tissues, complement system activation must be tightly controlled. The regulating complement control proteins, including CD59, also known as protectin, are present at a higher concentration in the blood plasma than the complement proteins themselves. The complement system is thought to play a key role in many diseases with an immune component, such as asthma and many autoimmune diseases, including SLE, IBD and multiple sclerosis. It is also becoming increasingly associated with neurological disease, such as Alzheimer′s disease and conditions such as spinal cord injuries.

MILLIPLEX® Complement Panel 1 Bead Panel is an 7-plex kit to be used for the simultaneous quantification of any or all of the following analytes in serum, plasma or culture supernatant samples: C2, C4B, C5, C5a, Adipsin/Complement Factor D, MBL and Complement Factor I.

The Luminex® xMAP® platform uses a magnetic bead immunoassay format for ideal speed and sensitivity to quantitate multiple analytes simultaneously, dramatically improving productivity while conserving valuable sample volume.

  1. Do the Human Complement Milliplex Panels cross-react with mice?

    1. In our testing, 8 mouse samples were tested in both Human Complement Panels, and only Factor B was detected. While we cannot definitively assert that there is no cross-reactivity, it is worth noting that the lack of signal could be attributed to low levels in the few samples tested. However, we did not observe any obvious cross-reactivity.

  2. Can the Human Milliplex Complement Panels HCMP1MAG-19K and HCMP2MAG-19K be used to measure Complement Factors in rabbits?

    1. The Human Complement Milliplex panels have not been tested with rabbit samples. There are no references available where this has been done. It is uncertain whether the kits can detect rabbit samples.

    1. We have not tested the assay for free C5a only. The antibody specificity information does not list this detail. The immunogen is only listed as purified human C5a.

  4. Do the Adipsin antibodies exhibit consistency between the Human Adipokine Panel 1 (HADK1MAG-61K) and Human Complement Panel 1 (HCMP1MAG-19K)? Is there a correlation in the sample data?

    1. Certainly, the Adipsin capture and detection antibodies are consistent across both panels. Furthermore, the sample data demonstrates a high correlation, with an R value of 0.99 and a slope of 0.7.
