过氧化氢是由分子氧代谢产生的活性氧类物质,既是一种细胞内信号传导信使,也是一种氧化应激来源。过氧化氢由细胞通过多种机制生成,如中性粒细胞和巨噬细胞内NOX介导的ROS产生(呼吸爆发),或者通过在线粒体呼吸期间由电子泄漏产生的超氧阴离子的歧化酶催化产生ROS。异常的过氧化氢产生将导致细胞氧化损伤以及哮喘、动脉粥样硬化、骨质疏松症和神经变性等疾病进展。

原則
过氧化氢检测试剂盒(荧光法)还可用于检测活细胞释放的过氧化氢或者由氧化酶活性产生的过氧化氢。该试剂盒利用过氧化物酶底物与过氧化氢反应,产生的红外荧光产物(λex = 640/λem = 680 nm)可用荧光酶标仪进行分析。 在pH 4-10范围内,荧光产物的生成不受pH影响,因此,该试剂盒优于需要在低pH溶液中测定过氧化氢的其他荧光试剂盒,后者的探针灵敏度降低。 红外荧光探针对于由生物样品自发荧光造成的测定背景不太敏感。该测定兼容高通量操作系统。
