Allgemeine Beschreibung

Das F2-Gen codiert Thrombin-Protein und ist auf dem humanen Chromosom 11p11.2 lokalisiert.[1]

Anwendung

Thrombin aus Humanplasma wurde:
  • in CaCl2 gelöst und zu einer Polyethylen-Glycol-Fibrinogen/Zelllösung zur Fibrinherstellung gegeben[2]
  • zur Polymerisation von Fibrin auf Mikrowellplatten verwendet[3]
  • als eine endogene Agonist-Proteinase für proteinaseaktivierte Rezeptoren: PAR1, PAR3 und PAR4 zur Stimulation von Zellen eingesetzt[4]

Biochem./physiol. Wirkung

Serinprotease, die selektiv Arg-Gly-Bindungen in Fibrinogen spaltet, um Fibrin und Fibrinopeptide A und B zu bilden.
Thrombin ist eine endolytische Serinprotease[5], die eine durch Gefäßschäden induzierte Produktion von Leukozyten, Thrombozyten, Endothelzellen und Mesenchymzellen stimuliert. Diese Aktion wird über einen G-Protein-gekoppelten Rezeptor vermittelt.[6] Es katalysiert die Spaltung von Fibrinogen an Arg−Gly-Bindungen, wobei Fibrin gebildet und die Fibrinpeptide A und B freigesetzt werden.[5] Die überwiegende Form von Fibrin in vivo ist das Zymogen Prothrombin (Faktor II), das in der Leber produziert wird. Prothrombin ist ein Gerinnungsfaktor, der in hohen Konzentrationen auf eine Krebserkrankung hindeutet. [5] Thrombin kann zur Spaltung von Fusionsproteinen eingesetzt werden. Die Spaltung von Fusionsproteinen kann bei einem Thrombin:Fusionsprotein-Verhältnis von 1:500 durchgeführt werden. [7]

Einheitendefinition

Die Aktivität wird in NIH-Einheiten ausgedrückt, die sich aus einem direkten Vergleich mit einem NIH-Thrombin-Referenzstandard ergeben.

Lyophilisiert aus Kochsalz-Natriumcitrat-Puffer, pH 6,5

Hinweis zur Analyse

Beim NIH-Assay werden 0,2 ml verdünntes Plasma (1:1 mit Kochsalzlösung) als Substrat und 0,1 ml der Thrombinprobe (stabilisiert in einer 1%igen gepufferten Albuminlösung) auf Basis einer modifizierten Biggs-Methode analysiert. Zur Bestimmung der Thrombinkonzentrationen werden nur Gerinnungszeiten im Bereich von 15-25 Sekunden verwendet.

Since thrombin solutions adsorb to glass, it is recommended to aliquot the solutions in plastic tubes and store frozen at -20°C or colder for long term storage.

Thrombin from any vertebrate species will clot the fibrinogen of virtually any other species.  Fibrinogen from any mammalian source will cross-react with thrombin from any mammalian source, but it is not well known how thrombin and fibrinogen from different animals differ in their sequence.  When any mammalian thrombin is injected into a different mammal, clotting will occur.
