Fibroblast growth factor 1 (FGF1) is an autocrine/paracrine regulator whose binding to heparan sulphate proteoglycans effectively precludes its circulation. Although FGF1 is known as a mitogenic factor, FGF1 knockout mice develop insulin resistance when stressed by a high-fat diet, suggesting a

Questions

1–2 of 2 Questions
  1. How many times the solution of heparin can be used after resuspension in water? And what are the storage conditions of this solution?

    1 answer
    1. There is no Product Information Sheet specific for product SE0027. There are other sodium heparin products from the porcine intestinal mucosa. The Product Information Sheet for H3393 states:
Storage/Stability: Store the powdered product at room temperature.
Heparin is a polysaccharide, which remains active in aqueous solution for up to 2 years at 2–8 °C, provided the solutions have been filtered through a 0.2 mm membrane. Alternatively, 0.06–1% benzyl alcohol may be added to solutions to prevent bacterial growth. Any trace of contaminating organisms in the solution will break down the heparin to use its sugars for nutrients, resulting in the degradation of non-sterile solutions.
Heparin solutions should not be autoclaved, because additional crosslinking of the sugars may occur at high temperatures. There is no need to freeze solutions of heparin; in fact, solutions of any polysaccharide of high molecular mass should not be frozen.

Other sources state that the pH should be between a range of pH 4 to pH 9.

  2. What is the molecular weight of this product? How many milligrams? sre0027-100KU

    1 answer
    1. The exact amount of material will vary per batch and will be based on the unit activity. For example, on the sample Certificate of Analysis, the specific activity is 206 units/mg. This means that for the 100KU pack size, there is 485 mg of material: https://www.sigmaaldrich.com/certificates/sapfs/PROD/sap/certificate_pdfs/COFA/Q14/SRE0027-100KUSLCQ0488.pdf

This product does not have an indicated molecular weight, however, per Merck Index, 11th ed., p. 735, #4571: Molecular weight of heparin is 6 – 30 KDa.
In addition, an analysis of 32 commercial samples of heparin sodium (U.S.P.) and sterile solutions of heparin sodium (U.S.P.) indicated that the average molecular weight was 9 – 12 KDa.
