Principio

When blood films are stained using Giemsa Stain, the nucleus and cytoplasm of white blood cells take on characteristic blue or pink coloration. The use of purified eosin and thiazine dyes minimizes lot-to-lot variation.

    1. Giemsa solution protocols recommend diluting with either water or a phosphate buffer. Common phosphate buffers are Weise or Sorensen's phosphate buffer. Depending upon personal preference, the pH used can vary from a low pH of 6.0 to a high of 7.2 In North America it is more common to use a Sorensen's phosphate buffer with a pH ranging from 6.8 to 7.2. A suitable catalog number for a pH 7.2 buffer is P3288. In other parts of the world slightly more acidic phosphate buffers are preferred.

  2. Hi does any lab use this stain for microscopy ( haematology) ? How is it working for you on fresh peripheral blood films ?

    1. Giemsa solution is commonly used for hematology, histology, and parasitology labs. Peripheral blood smears and bone marrow smears are routinely stained in hematology labs. It is always preferred to use fresh peripheral blood smears or bone marrow smears when possible. The colors are more vibrant when the peripheral blood smears are stained fresh.

  3. Is there a white paper available for buffer selection for Giemsa staining?

    1. Generally, no white papers are available. Giemsa formulations vary in pH, typically ranging between pH 6.4 to 6.9. Quality standards for stained slides differ across institutions and individuals, with varying preferences for pH buffers.
