Description générale

Krebs-Ringer Bicarbonate Buffer acts as a salt solution and plays an essential role in tissue culture. A salt solution provides the cells with water and inorganic ions. It also helps in maintaining the osmotic balance and ideal pH in the medium. This bicarbonate buffered isotonic saline contains magnesium, potassium, sodium, and phosphate ions.

Application

Krebs-Ringer Bicarbonate Buffer has been used:

  • as a wash buffer for the preparation of mice primary adipocytes and lipolysis assay[1]
  • in the pre-perifusion for islet viability and functionality studies[2]
  • in pituitary bioassay[3]

Quantité

Formulated to contain 9.5 grams of powder per liter of medium.

Reconstitution

Supplement with 1.26 g/L sodium bicarbonate.

  4. Product K4002 - Krebs-Ringer Bicarbonate Buffer, is without calcium chloride and sodium bicarbonate. In the reconstitution steps, you advice to add 1.26g sodium bicarbonate. Should we also add some calcium chloride? in which amount?

    1. Product K4002 - Krebs-Ringer Bicarbonate Buffer does not contain calcium. The addition of calcium is not required for osmolarity.  If you need to have calcium, you can add calcium chloride between 1.25 to 2.5 mM.  You may want to check the osmolarity and make sure it is not too high.Kreb Henseleit Buffer has 1.25 mM calcium chlorideKreb's Buffer has 2.5 mM calcium chlorideKreb' Ringer Superfusion solution contains 2.0 mM calcium chloride.Please see the attached document for different Krebs' buffer recipes.
