Description générale

stable jusqu'à six mois s'il est conservé au réfrigérateur à un pH inférieur à 7. Si un pH supérieur à 7 est nécessaire, il convient d'ajuster le pH juste avant utilisation.

Application

Le chlorhydrate du fluorure de 4-(2-aminoéthyl)benzènesulfonyle (AEBSF) a été utilisé :
  • comme inhibiteur de protéases dans des échantillons de plasma pour éviter la dégradation de la ghréline acylée (AG).
  • comme inhibiteur de sérine protéinases de venin de serpent (SVSP) dans un essai d'inhibition de SVSP ou un essai sur l'AEBSF pour tester l'implication des SVSP dans l'activité de la coagulation due au fibrinogène.
  • comme constituant d'un tampon de lyse NP-40 pour remettre en suspension des culots cellulaires afin de préparer des lysats cellulaires.

Les concentrations recommandées pour l′emploi sont comprises entre 0,1 et 1 mM.

Actions biochimiques/physiologiques

L'AEBSF est capable d'inhiber l'acétylcholinestérase (AChE).
Le chlorhydrate du fluorure de 4-(2-aminoéthyl)benzènesulfonyle (AEBSF) est un inhibiteur irréversible des sérine protéases. Ses constantes d'inhibition sont proches de celles du fluorure de phénylméthanesulfonyle (PMSF) et du diisopropylfluorophosphate (DFP). Il a été démontré que l'AEBSF inhibe la trypsine, la chymotrypsine, la plasmine, la kallikréine et la thrombine. Comme option de remplacement du PMSF ou du DFP, l'AEBSF offre une plus faible toxicité, une meilleure solubilité dans l'eau et une plus grande stabilité dans les solutions aqueuses. L'AEBSF peut être utilisé en culture cellulaire à des concentrations allant jusqu'à 0,25 mM.

If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

    1. It is highly soluble in water and in DMSO, at 50 mg/mL (approximately 200 mM), yielding a clear colorless solution. AEBSF may be used in cell culture medium at concentrations up to 0.25 mM.

    1. Solutions in water are slightly acidic and retain inhibitory activity for up to six months when stored refrigerated. Solutions at pHs above 7 are less stable.  Stock solutions should be stored at a pH less than 7. If a final pH of greater than 7 is required, the pH adjustment should be done shortly before use.
