Os ácidos graxos livres, também conhecidos como ácidos graxos não esterificados, circulam no plasma ligados a albumina. A maioria dos ácidos graxos livres é derivada de fontes alimentares ou é mobilizada do tecido adiposo. Os níveis de ácidos graxos livres plasmáticos são elevados em muitas doenças relacionadas à obesidade e podem contribuir para a resistência à insulina nos tecidos periféricos. Quadros como sepse e tumores que produzem hormônios lipoativos também podem estar associados a níveis elevados de ácidos graxos livres.

Alterações no metabolismo de ácidos graxos causam a doença hepática gordurosa.[1] O acúmulo de ácidos graxos livres nos miócitos leva ao risco de disfunção cardíaca.[2] A β-oxidação elevada de ácidos graxos está envolvida na isquemia cardíaca.[2]

Aplicação

O kit de quantificação de ácidos graxos livres foi usado:
  • para medir a hidrólise de lipídios por determinação de ácidos graxos livres em amostras digeridas.[3]
  • para medir o nível de ácidos graxos livres no plasma e no soro.[4][5]
  • para quantificar o nível de ácidos graxos livres no tecido hepático.[6]

Adequação

Adequado para detecção de ácidos graxos livres de cadeia longa em diversas amostras biológicas, como soro, plasma e outros fluidos corporais, alimentos, meios de crescimento, etc.

Princípio

Nesse kit, a concentração de ácidos graxos (C8 e mais longos) é determinada por um ensaio enzimático acoplado, o que resulta em um produto colorimétrico (570 nm)/fluorométrico (λex = 535/λem = 587 nm) proporcional aos ácidos graxos presentes.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   3. Is there a step in the process where samples can be preserved for future analysis using the kit?

    1. Yes, samples can be preserved for future analysis using the kit by aliquoting and freezing them if needed for multiple uses. However, it's recommended to use fresh samples whenever possible to ensure accurate results. Dried lipids can be stored in 200 µl of Fatty Acid Assay Buffer at -80°C. In case of lipid precipitation, re-dissolve the lipids by sonication. However, the supplier does not specify the maximum duration for which the samples stored at -80°C will remain stable. Storing samples at -80°C may impact their stability, potentially resulting in lower than expected readings. Therefore, it is recommended to use fresh samples for this assay whenever possible to ensure accurate results.
      If frozen samples are used, they should be snap frozen in liquid nitrogen immediately after collection. These samples cannot undergo multiple freeze-thaw cycles and must be stored for relatively short periods. It's important to note that stability studies on different sample types have not been conducted, so there is no advice available regarding the timeframe for measuring frozen samples.

  4. Can MAK044 quantify only albumin-bound free fatty acids (FFA), or does it also quantify unbound FFA?

    1. The kit specifically detects unbound free fatty acids in the samples and does not detect any albumin-bound free fatty acids.

  5. What are the recommended storage instructions for the Enhancer reagent and unused Assay Buffer in the MAK044 Free Fatty Acid Quantitation Kit?

    1. The enhancer can be aliquoted and stored at -20°C, while the assay buffer can also be stored in aliquots at -20°C to avoid multiple freeze-thaw cycles.

  6. I would like to quantify butyric acid in bacteria culture supernatant. Does this MAK044 Free Fatty Acid Quantitation Kit suitable or is there a kit you can recommend. Thank you.

    1. Butyric acid is a short-chain fatty acid. This kit is suitable for detecting the long-chain free fatty acids in various biological samples, such as serum, plasma, and other body fluids, food, growth media, etc. The Short Chain Fatty Acid Kit, prod number SBR0030, offers a potential analytical option. This kit is suitable for detection of several SCFAs, including Butyric, by HPLC, LC-MS, and GC-MS. See the link below to the technical bulleting should these methods be suitable:

  7. Can this kit be used to analyse NEFA in plasma using the Pentra C400 instrument?

    1. This kit is suitable for detecting nonesterified fatty acids in plasma (NEFA). However, suitability with Pentra C400 instrument has not been determined. This application must be determined by the end-user.

  8. Can these kits be used with serum samples from mouse?

    1. Yes, this kit can be used on mouse serum samples. We generally recommend that fresh serum samples be used. Frozen samples can probably be used. However, the successful use of frozen samples depends on the efficiency of freezing them and the amount of time they have been frozen. If previously frozen serum samples are used, you may want to check the performance of the kit with fresh vs. frozen samples.
