General description
Thyreotropes Hormon/Thyreoidea-stimulierendes Hormon (TSH) gehört zur Familie der Glykoproteinhormone. Dieses Glykoprotein wird von thyreotropen Zellen des Hypophysenvorderlappens erzeugt. Es zirkuliert in freier Form und besitzt eine kurze Plasma-Halbwertszeit.
Application
Das thyreotrope Hormon aus der Rinderhypophyse wird wie folgt verwendet:
- zum Induzieren der cAMP-Produktion in orbitalen Fibroblasten;
- als Bestandteil von Dulbeccos Modified Eagle Medium (DME) H-21:F-12;
- für einen TSH-Stimulationstest zur Beurteilung der Schilddrüsenfunktion nach induziertem Euthyroid-Sick-Syndrom (ESS);
- zur Untersuchung der Beteiligung des thyreotropen Hormons an der photoperiodischen Signaltransduktion bei Mäusen.
Biochem/physiol Actions
Thyreotropes Hormon/Thyreoidea-stimulierendes Hormon (TSH) unterstützt die Regulierung der Schilddrüsenfunktion. Bei Kindern spielt das TSH eine wichtige Rolle für die normale kognitive und neuronale Entwicklung. Zirkulierendes TSH kann Rezeptoren auf der Zelloberfläche in der Schilddrüse binden. Dieses Hormon veranlasst die Schilddrüse, Thyroglobulin und die Schilddrüsenhormone zu produzieren. Das TSH kann bei Menschen mit Schilddrüsenkrebs auch die Aufnahme von Iod (131L) fördern. Außerdem kann es die Sekretion des vaskulären Endothelwachstumsfaktors (VEGF) und die Angiogenese in Zelllinien mit Schilddrüsenkrebs fördern.
Zweikettiges Glykoproteinhormon. Die α-Kette ist nicht aktiv, biologische Spezifizität wird eher mit der β-Kette in Verbindung gebracht. Es aktiviert Adenylatcyclase in der Schilddrüse und stimuliert die Iodaufnahme sowie die Thyroxinsynthese und -freisetzung. Goitrogen.

Without IU, what concentration in mg/ml is typically used in thyroid cell culture?

Historically, the activity of this product was determined to have a range of 1.5 - 2.5 IU/mg protein. This product has not been screened for activity for a number of years. There are publications citing the use of bovine TSH in uM concentrations. Please see the link below to review an example:
